We examined these associations in the National Alzheimer's Coordinating Center cohort and compared glucosamine with other supplement records captured in the same medication fields. The estimate for progression to primary AD dementia was in the same risk direction as the original finding, but similar estimates were observed for multivitamin/broad vitamin and calcium/vitamin D records. Among participants with dementia, glucosamine records were not associated with higher adjusted mortality, although death-data limitations reduce certainty. These findings do not address the experimental mechanism, but show that a glucosamine record alone does not establish a glucosamine-specific clinical effect.",

"abstract": "Right ventricular (RV) trabeculation may reflect adaptation to loading conditions and imprinting of early developmental processes, yet its genetic determinants and clinical relevance in adults remain poorly understood. In contrast to the left ventricle, the RV has distinct developmental origins, geometry and loading conditions, suggesting chamber-specific mechanisms of trabecular remodelling. Using deep learning-based image segmentation and fractal dimension analysis, we quantified RV trabecular complexity in diastole and systole in 48,118 UK Biobank participants with genetic data. RV trabecular morphology was associated with systolic function, as well as cardiometabolic and respiratory conditions. Genetic analyses across the allele frequency spectrum identified 52 common loci and 45 genes with a burden of protein-altering variants, implicating sarcomeric function, cytoskeletal organisation, and early cardiac patterning. Although many loci showed shared effects across both ventricles, we also identified RV-specific genetic associations linked to congenital heart disease and respiratory phenotypes. "abstract": "Sample preparation remains a barrier for decentralized, swab-based molecular testing of tuberculosis (TB). Extraction-free workflows offer a simpler alternative, but systematic benchmarking against standard methods is lacking. We evaluated five novel lysis devices, BLINK Shaker Prototype, nPOC-BB, SPS-1, Truelyse, and Thermolyse, against a heat- and bead-beating reference method using contrived M. tuberculosis-spiked tongue and sputum swabs. The primary outcome was lysis efficiency, measured as the relative DNA recovery compared with the reference workflow. Secondary outcomes included nuclease inactivation, biosafety, and usability. In the reference buffer, lysis efficiencies ranged from 51-63% to 95-154% on tongue swabs and 12-54% to 280-644% on sputum swabs across the five devices. In proprietary buffers, performance varied more widely, with lysis efficiencies of 2-4% to 64-80% on tongue swabs and 1% to 94-398% on sputum swabs. Complete biosafety inactivation was achieved by three devices; two showed residual growth (<0.02%). "abstract": "Background Polyendocrine Metabolic Ovarian Syndrome (PMOS), formerly known as Polycystic Ovary Syndrome (PCOS), is a prevalent endocrine disorder with high rates of undiagnosed cases globally. As PMOS is frequently characterised by oligo-anovulation, the absence of the characteristic rise in basal body temperature typically seen in ovulatory cycles may serve as a physiological marker for the condition. Objective This study aimed to assess the feasibility of using machine learning to identify individuals with PMOS from temperature data collected by a body-worn device. Methods We used data from 387 users of a vaginal temperature monitor (OvuSenseTM) who responded to a questionnaire. "abstract": "Objectives: To develop a multidimensional framework for assessing organisational and system readiness for federated health-data services, and to report its formative application across three heterogeneous UK ecosystems. Methods: The Health Data Readiness Level (HDRL) framework was developed from a structured landscape review of 56 maturity and readiness frameworks, first-principles requirements analysis, artificial-intelligence-assisted synthesis with human source verification, and stakeholder refinement. It comprises 64 indicators in eight domains and five ordered levels. Formative application examined three heterogeneous UK health-data research ecosystems using documentary evidence, professional stakeholder input, workshops, a structured right-of-reply process, cross-case calibration, and two illustrative research use cases. Analysis was descriptive; the application was not designed as psychometric validation or a league table. Results: All 64 indicators were scoreable in each case. "abstract": "Passive physical activity is increasingly used as a digital-phenotyping proxy for mood, assuming a stable relationship across people and time. We tested this assumption using daily mood and activity data from 1,072 individuals (121 women, 951 men; 13,909 person-days) in the Juli app. The within-person activity-mood association was stronger in women than men (slope difference -0.194, p = 0.019). Within women, it varied across the menstrual cycle (p = 0.022): absent in the early luteal phase, significant in all other phases, and largest in the late luteal phase (+0.46). A male pseudo-cycle control showed no such modulation (p = 0.974), confirming the effect is cycle-specific rather than a general temporal pattern. Accounting for cycle phase improved out-of-sample mood prediction in women in 75% of cross-validation splits. Digital phenotyping should account for sex and menstrual cycle phase to avoid biased predictions and enable personalized recommendations for women.",

The level of concordance on fertility desires was assessed using Kappa Coefficient. Principal component analysis (PCA) was applied to assess the internal consistency and dimensionality of household decision-making and IPV attitude items; items within each construct were subsequently summed to generate categorical empowerment scores used in the regression analyses. Relative risk ratios were estimated for perceived empowerment on spousal concordance on fertility desires using multinomial logistic regressions. Results The study found a moderate level of spousal agreement on fertility desires (Kappa=0.51). "abstract": "Women engaged in informal cross-border trade in Southern Africa experience frequent mobility, economic pressure, and exposure to new social environments that may increase vulnerability to risky sexual behaviour and HIV. Limited evidence exists on how pre-migratory, migratory, and post-migratory factors interact to shape sexual risk among this population. This study examined the prevalence and determinants of risky sexual behaviour among women informal cross-border traders at COMESA Market in Lusaka, Zambia. A cross-sectional survey was conducted with 499 women informal cross-border traders. A composite risk score was used to classify respondents as \"low risk\" or \"high risk\" for sexual behaviour. Descriptive statistics, chi-square tests, and multivariate binary logistic regression were used to investigate associations between selected covariates and risky sexual behaviour, guided by migration and vulnerability theory. The majority (67.7%) of respondents were classified as high-risk. In multivariate analysis, women who used personal savings for start-up capital were 73% less likely to be high-risk compared to those relying on nuclear family capital (aOR = 0.27, 95% CI: 0.10-0.73).
